C57BL/6-TgH(mito-cpYFP)小鼠模型的构建及其发育过程中自由基分布规律的研究

  mito-cpYFP是近年来新发现的自由基探针,它的优点在于可以定点的表达在细胞线粒体内,被线粒体内呼吸链的电子泄露产生的超氧自由基氧化而产生荧光,具有可逆性,可以原位、实时的反映出超氧自由基在生物体内的分布。本实验将mito-cpYFP的基因序列转入C57BL系小鼠体内,拟构建mito-cpYFP小鼠模型,通过对构建出的新品系小鼠进行DNA及mRNA的鉴定,结果均为阳性。此外,利用小动物荧光成像系统观察并分析得到,在C57BLTgH(mito-cpYFP)小鼠出生一周内,其体内超氧自由基的浓度每日呈先上升后下降的趋势,并在11:30-12:30达到最高,且在一周之内超氧自由基的浓度也是先增长后下降。进一步观察可以知道,该小鼠的腹部是其自由基最开始的主要聚集处,随着年龄的增长,腹部的超氧自由基逐渐延腹白线向上延伸。因此,可以判断mito-cpYFP小鼠模型构建成功,且获得了幼鼠发育过程中超氧自由基在体内的分布状况,即延腹白线分布。
