BackgroundInositol 1,4,5-trisphosphate receptor (IP3R) is abundant in the vascular smooth muscle cells "SMCs" and IP3R form tetrameric channels in endoplasmic reticulum membranes that conduct Ca2+ in an Inositol 1,4,5-trisphosphate (IP3)-sensitive manner.IP3-mediated Ca2+ release is regulated by [Ca2+]i,with positive and negative feedback acting in a bell-shaped manner with peak Ca2+ release activity at 300 nM Ca2+.Nitric oxide (NO) is a negative regulator of vascular SMC proliferation,which its antiproliferative effect is mediated via guanosine-3',5'-cyclic monophosphate (cGMP) and cGMP dependent protein kinase (PKG).
