MiR482 Negatively Regulates the Expression of CC-NBS-LRR Resistance Genes in Tomato

  NBS-LRR disease-resistant gene family is the most representative family of resistance genes.Previous studies have shown that miR482 targetes NBS-LRR mRNA sequences and regulates the immune response in plants.To study the interaction between different miR482 family members and their target genes in tomato, we downloaded the sequences of three miR482 family members (miR482a,miR482b, miR482c) from the miRBase, following by the prediction of their target genes.According to the predicted results and the relative reports, we selected Solyc02g036270.2.1 (CC-NBS-LRR) to study the interaction relationship between the target gene and the three miR482 family members.We assayed the changes of expression of miR482 family members and their target gene by quantitative real-time PCR after Phytophthora infestans induced for different time respectively.The results of quantitative real-time PCR and correlation analysis indicated various miR482 family members negatively regulate the expression of their common target gene on different levels.The result further revealed the interaction between the miRNAs and their target genes, providing a new theoretical basis for the breeding of tomato disease resistance.
