PCAF Regulates the Inflammatory Response in the Development of Diabetic Nephropathy

  Diabetic nephropathy is a common diabetic microvascular complication.Recently, several studies have demonstrated that the changes ofhistone modifications are linked to the development of diabetic nephropathy, however, the exact mechanism remains unclear.In the present study, we observed significant increases in acetylated histone H3 with up-regulated of PCAF, a histone aectyltransferase, in the renal cortex of db/db mice compared to the control mice.In human proximal tubule epithelial HK-2 cell, PCAF knockdown deceases the inflammatory factors in the both mRNA and protein levels, including vascular cell adhesion molecule 1 (VCAM-1), intercellular adhesion molecule 1 (ICAM-1), and monocyte chemoattractant protein 1 (MCP-1).
