昆虫的几丁质酶家族group I主要负责蜕皮过程中表皮几丁质降解,被认为是农药设计的潜在靶标.通过分析亚洲玉米螟几丁质酶OfChtI催化域的晶体结构,发现其活性口袋底物结合裂缝处存在一系列芳香族氨基酸,提供其与底物结合的关键疏水作用,同时也是基于结构进行选择性抑制剂筛选的重要靶点.基于以上发现,通过对比底物几丁三糖化合物结构、电荷性质对商品化合物数据库进行虚拟筛选,获得了其抑制剂TP1,TP2.
目前对生物大分子的定量研究主要采用“绝对定量”和“相对定量”的方式。但是,由于研究对象—生物体的多样性、动态性和复杂性,导致了以往研究所采用的定量体系已经不能满足于大数据时代分子生物学研究的需要。对于原始数据的产出,分子生物学研究亟需在方法学上建立新的理论体系,新的生物大分子定量规范和新的生物样本定量规范需要重新建立。在新的生物定量模式下开展定量分子生物学研究将是分子生物学的发展趋势。
