Objective Detect the effects of CSPGs on proliferation and migration of OPCs as well as the mechanisms of differentiation effects of CSPGs on OPCs.Explore the mechanisms of hypomyelination in preterm white matter injury.Methods OPCs from neonatal SD rats brain were cultured in vitro.MTS and EdU incorporation methods were used to detect the differences of proliferation rates among CSPGs groups and control group.Transwell migration assay was administered to observe the effects of CSPGs on migration of OPCs.Western blot and qPCR were used to detect the expression differences of β1-integrin and its mRNA levels between CSPGs groups and contral group on 3 days and 5 days after differentiation.After coating the plates with 9 different combinations with CSPGs and laminin at 3 concentration levels.We observed the morphological changes of OLs by immunofluorescence at 5 days after differentiation.The proportions of 4 different phenotypes of oligodendrocytes among groups were calculated.The main effects and interaction between CSPGs and laminin on OPCs differentiation were analyzed.
