Objective To investigate the correlation between cyclosporine blood concentration of heart transplant patients and PXR gene polymorphism.Methods Time flight mass spectrometry (MassARRAY) was applied to detect the genes of 59 cases of patients with stable heart transplant recipients of PXR, AxSYM automated chemistry immune analyzer was used to test the serum concentration of cyclosporine, and then the influence of PXR single nucleotide polymorphism (SNP) on cyclosporine trough blood concentration was analyzed by statistical methods.Results The minor allele frequency (MAF) in 8 tags of PXR single nucleotide polymorphism (SNP) in our study was similar with the MAF in NCBIs dbSNP database of Chinese peoples.For the 59 patients with stable heart transplant, there is no statistical significance in the cyclosporine blood concentrations among the genotype groups of these seven PXR Tag SNP rs3814056T>G, rs7643645A>G, rs11917714C>T, rs2276705C>A, rs2472681T>C, rs2472682C>A, rs4440154C>T.Only PXR rs1523127 (C 24381 A) AA patients serum concentration and corrected concentration are obviously lower than CA patients, whereas CA patients concentration are lower than CC patients, whose difference is significant in statistics(P<0.05).Conclusion: This study showed that in 8 PXR Tag SNP, there is correlation between PXR rs 1523127 (C 24381 A) genotype and cyclosporine blood concentration of the stable patients with heart transplant, displaying that this SNP may take effect in individual cyclosporine application.
