Objective Endothelial-monocyte-activating polypeptide Ⅱ(EMAP Ⅱ)has been shown to inhibit tumors by targeting vascular endothelium. However, the effect of EMAP Ⅱ on glioma cell of treating brain glioma has not been precisely elucidated. The present study was performed to examine whether EMAP Ⅱ inhibit glioma growth by inducing rat brain glioma C6 cells apoptosis. Methods The viability of rat C6 glioma cells was detected by MTT assay. The Nuclear characteristic changes of apoptosis were visualized by Hoechst DNA staining.Flow cytometry was used to analyze the apoptotic proportion and the change of mitochondrial membrane potential(MMP)of C6 glioma cells. The activity of caspase-3 and caspase-9 were tested by respective activity assay kit.Western blot assay was used to test the protein expression of caspase-3,caspase-9,reactive oxygen species(ROS),Bcl-2,Bax and Apaf-1.
