Although the involvement of Glu,GluR,AVP,AVPR in anxiety have been investigated in previous studies respectively,but the effects of interactions between them on anxiety were still unclear.In present study,the agonist and antagonist either for NMDA receptor and AMPA receptor,as well as AVP and V1aR receptor antagonist were given to Babl/cj mice by intracerebroventricular microinjection and the levels anxiety-like behavior was investigated by open field test and elevated plus maze test.The result showed that Babl/cj mice displayed high levels of anxiety-like behavior compared to C57 mice.The NMDA receptor antagonist (MK-801),AMPA or V1aR blocker (SSRI49415) have significant anxiolytic effect.AVP in combination with MK-801 or SSRI49415 together with DNQX also decreased the levels of anxiety.These results suggest that the expression of anxiety-like behavior in BALB/CJ mouse possibly results from the coordination among the glutamate,NMDA receptor,AMPA receptors,AVP and V1aR.With MK-801,AMPA or SSRI49415 having a significant anxiolytic effect,we hypothesized that the development of anxiety behavior in BALB/CJ mice may be due to the alterations of NMDA receptor and V1aR,as well as decreased activity in AMPA receptor.The results also suggest that AMPA receptors play an anxiolytic effect through inhibition of the AVP and V1aR.AVP alters the levels of anxiety through binding to V1aR.In addition,the role of AVP also may be through the activation of NMDA receptor via binding to V1aR.
