胃癌与饮食、感染、环境、遗传等因素关系密切,属于一种多基因疾病.该病致死率较高,全世界每年约有647,000人因胃癌死亡,位居癌症死因的第2位.近年来,尽管胃癌发病率在全球呈下降趋势,但在中国、日本和韩国等地区仍维持较高的发病率.该病已严重影响到人民群众生命健康,开展胃癌防治研究已成为当前我们面临的重要课题之一.前期我们课题组在体临床观察发现幽门螺杆菌感染相关胃癌存在CD14基因型变化差异,CD14基因多态性可能与胃癌易患正相关,由此课题组在离体利用靶向性CD 14shRNA慢病毒载体,转染胃癌细胞SGC-7901后,观察细胞增殖、凋亡及细胞周期变化,用Real-time PCR和Western-blot分别检测正常对照组、阴性对照组和CD14shRNA慢病毒载体组的CD14、TNF-α、IL-2的表达,进一步探讨RNAi抑制CD14表达对胃癌的作用,以期为胃癌防治提供新的思路.
