目的了解福州市2006年-2012年流行的乙型流感病毒血凝素基因变异特点及种系进化分布。方法采用狗肾传代细胞培养分离流感病毒,选取部分乙型流感病毒株进行血凝素基因HA1区核苷酸序列测定并推导出其氨基酸序列,然后用DNASTAR MegAlign序列分析软件进行基因种系进化特性分析。结果 2006年-2012年福州市人群中同时流行着乙型Yamagata系和Victoria系毒株。分离的Yamagata系毒株与B/Florida/4/2006比较,在血凝素基因HA1区发生3个~7个氨基酸替换。分离的Victoria系毒株与B/Brisbane/60/2008比较,在HA1区发生5个~7个氨基酸替换,在197位增加一个糖基化位点。结论福州市人群中流行的乙型流感病毒与Yamagata系和Victoria系代表株相比基因特性已经发生了进一步改变。
Objective To understand the genetic variation of the hemagglutinin gene and the phylogenetic distribution of influenza B virus circulating in Fuzhou from 2006 to 2012. Methods The influenza virus was isolated from the passaged cells of canine kidney and selected part of influenza B virus strains to determine the amino acid sequence of HA1 gene of hemagglutinin gene and then deduced the amino acid sequence of HA gene. Then DNASTAR MegAlign sequence analysis software was used to analyze the germline evolution characteristics analysis. Results From 2006 to 2012, both strains of Yamagata and Victoria strains were prevalent in Fuzhou. The isolated strain of Yamagata had a 3-7 amino acid substitution in the HA1 region of the hemagglutinin gene compared to B / Florida / 4/2006. Compared with B / Brisbane / 60/2008, the isolated strains of Victoria strain had 5-7 amino acid substitutions in the HA1 region and a glycosylation site at 197. Conclusions The genetic characteristics of influenza B virus circulating in Fuzhou population have been further changed compared with Yamagata strain and Victoria strain.
