Objective: the study investigated the effects of intra-nucleus accumbens (NAc) galanin and its receptor antagonist galantide injection on antinociception in mononeuropathic rats.Methods: the present study was performed in rats with experimentally induce mononeuropathy after left sciatic nerve loose ligation.The effects of intra-NAc injection of galanin and galantide on hindpaw withdrawal latency(HWL) to noxious thermal and mechanical stimulation were observed.Results: (1) 7-14 days after left sciatic nerve ligation, significant bilateral decreases on HWL to noxious thermal and mechanical stimulation.(2) Intra-NAc injection of 0.05, 0.1 or 0.5nmol of galanin, induce dose-dependent increases on HWL to noxious thermal and mechanical stimulation.(3)The galanin-induced antinociceptive effects are blocked by following intra-NAc injection galantide.Conclusion:galanin and galanin receptors in NAc were involved on antinociception in rats with sciatic nerve ligation.
