Objective To investigate the expression of Moloney leukemia virus 10(Mov10)in hepatocellular carcinoma and its biological functions.Methods The eukaryotic expression plasmid pCMV-Mov10-GFP was transfected into the HepG2 cells and then selected by G418.The mRNA and protein expression of Mov10 was measured by qPCR and Western blotting respectively.Immunofluorescence assay was used to detect the location of Mov10.The proliferation and colony-formation ability was measured by MTS assay and plate colony-formation assay.Results Our study indicated that the expression level of Movl0 in hepatocellular carcinoma was significantly higher than that in healthy liver.The same tendency was found in hepatoma cell lines compared with normal liver cell lines.We successfully established HepG2-Mov10 Cells which stably expressed Mov10 protein.HepG2-Mov10 cells had stronger ability of invasion and significantly higher proliferative rate than HepG2 cells (P < 0.05).Colonyforming efficiency was (56.7 ±17.3)%,(38.3 ±12.1) %,(31.3 ±8.1) % respectively in HepG2-Mov10,HepG2-GFP and HepG2 cells,and the HepG2-Mov10 showed significantly higher colonyforming efficiency than the ot her two cell lines (P < 0.05).Conclusion The expression of Mov10 in hepato cellular carcinoma was significantly higher than that in healthy liver.Stable over-expression of Mov10 protein promotes cell proliferation,colony formation and invasion.
