Laser scanning confocal microscope (LSM) have increasingly become routine laboratory facilities in the field of biology.Single-photon LSM has high resolution,sensitivity,and flexibility and is widely used in almost every biological laboratory.In our field of neurobiology,the two-photon laser scanning microscope achieves an efficient excitation depth within brain tissue using femtosecond lasers in the infrared spectrum.Combined with the high detection sensitivity of photomultipliers,this microscope is ideal for intravital imaging.Carl Zeiss has released an optical sectioning microscope that challenges the traditional meaning of a fluorescent microscope,the Light Sheet Z.1.The Light Sheet Z.1s illumination and imaging optics are placed vertically and it uses a cylinder to hold the sample.This microscope can image large samples like Zebrafish,Drosophila embryos,marine organisms,plant organs,and simultaneously gather single cell imaging data in vivo.The Light Sheet Z.1 delivers unmatched speed in volume imaging,the lowest photo-bleaching and photo-toxicity,and simultaneously allows for multi-photon microscopy for an in-depth look.
