  干扰素调节因子4(IRF4)通过转录调控细胞分化核心转录因子及分泌型细胞因子表达,从而介导体内多种天然免疫细胞如树突状细胞和巨噬细胞,辅助性T细胞如Th2, Th9,Th17,和调节性T细胞如FOXP3+Treg等不同细胞亚型的分化与功能。目前,IRF4在不同免疫细胞亚群中是如何行使其特定转录功能的分子机制尚不清楚。我们发现在TCR刺激条件下,CD4+T细胞中IRF4蛋白K123位点发生单甲基化,该位点的突变直接导致IRF4丧失转录激活其靶基因的调节活性。
