Temporal lobe epilepsy(TLE)is the most common type of epilepsy and is often medically refractory.Here we report pharmaco-genetical approaches that allow cell-type specific inhibition of neuronal excitability and control of TLE.Pharmaco-genetical activation of hippocampal parvalbumin neurons(a sub-type of GABAergic neurons)in seizure focus,using viral expression of the modified muscarinic receptor hM3Dq,attenuates both acute seizure model and chronic spontaneous epilepsy model with no obvious side effects.This anti-epileptic effect is mediated by GABAa receptor-mediated inhibition on pyramidal neurons.Meanwhile,directly pharmaco-genetical inactivation of pyramidal neurons,using viral expression of the modified muscarinic receptor hM4Di,produces similar anti-epileptic effect and further delays kindlinginduced epileptogenesis.These results indicate that pharmaco-genetical seizure attenuation may be a novel promising approach to treat refractory TLE with minimizing disruption of normal circuit function.
