Advantages of new dosage form like nanoparticles and microspheres in drug delivery system include improving water solubility,accumulating region of interest and enhancing antitumor efficacy,while reducing systemic toxicity.Liposomes,polymeric nanoparticles and microspheres have been developed to carry different drugs including anticancer agents,siRNA,and antiangiogenic agents.The present presentation provided an update on the Food and Drug Administration approved cancer nanomedicines and focused on those like liposomes,micelles and microspheres that have come into an advanced stage of clinical trials.Furthermore,the topic summarized the challenges from the development of new dosage forms land then shared the R&D process of the new dosage forms including nanoparticles,extended-release pellets of dry suspension,osmotic pump preparations,gastric retention preparations and oral rapid released drug system in our group.The advantages of extended-release pellets of dry suspension as drug carriers include enable formulation of solid sustained released dosage form,providing uniform drug absorption and reducing local irritation,improving palatability and flexibility of administration.Resulting from drug release profiles uninfluenced by medium composition and hydrodynamics,osmotic pumps preparations had many advantages,such as reducing side-effect,mitigating the food effect and exhibiting comparable in vitro/in vivo drug release.Control of placement of carriers in a specific region of the GI tract was suitable for drugs with limited absorption window in or unitability in the GI tract.Oral dissolvable film(ODF) is more and more becoming light-spot of oral rapid released drug systems.ODF has been reported to enhancing patient compliance by improving ease of administration and by not requiring water,which is easily adapted to the needs of individual patients or patient populations.
