Background The aim of the study is to establish the method to culture and induce human umbilical cord derived mesenchymal stem cells (hUC MSCs) to differentiate into cardiomyocytes in vitro.Methods hUC MSCs were isolated and purified from the umbilical cord of healthy full-term delivery fetus after the conset of puerpera by adhering to the culture dishes.The three passage of hUC MSCs was induced by different concentrations (5 μmol/L, 10 μmol/L, 20 μmol/L, 30 μmol/L and 40 μmol/L) of 5-azacytidine (5-aza) for four weeks,and then cultured in new medium.Control group was cultured in complete medium.Cells growth morphology was observed by convert microscope during the whole process.hUC MSCs surface antigens of passages 3 were measured by flow cytometry.Myocardium specific cardiac troponin Ⅰ (cTn Ⅰ) was examined with immunohistochemistry 4 weeks after induction.
