  miRNAs have been shown to be dysregulated in many types of hurnan cancer, including breast cancer.In particular,miR-21 was found to be significantly upregulated in several studies of human breast cancer tissues; this dysregulation was also found in ovarian and colorectal cancer.We have used miRNA microarrays from Exiquon to compare expression between normal mouse mammary glands and mouse mammary tumors induced by overexpression ofATF3.RNA from 4 individual tumors was compared to RNA from matched normal mammary glands, and duplicate arrays (3tumors) or a single array (one tumor) were hybridized following the manufacturers protocol.Data was processed with the LIMMA package in BioConductor/R.miRNAs were considered differentially expressed if the adjusted P-value (method of Benjamini and Hochberg) was <5 × 10-4 and expression was altered by at least 2-fold in at least three of the four tumors examined.Using these criteria, 14 up-regulated and 15 down-regulated miRNAs were identified.
