Background,Motivation and Objective Targeted contrast enhance ultrasound allows for the opportunity to visualize and track changes in molecular signalling pathways in relation to disease progression and response to therapy.To date,molecular ultrasound imaging using targeted microbubbles(MBs) have demonstrated selective localization to sites of angiogenesis,inflammation,and intravascular thrombus formation.However,the traditional approach has been to link biomolecules through interactions such as biotin-streptavidin noncovalent binding,which are not suitable for clinical use.Here we introduce a new strategy of MB targeting using a covalent,highly selective and rapid bioorthogonal coupling reaction:the inverse-electron-demand Diels-Alder reaction between tetrazine and trans–cyclooctene(TCO).
