Context: At present, The most often identifiable cause of POF is an autoimmune disorder of the follicular apparatus with a genetic cause established less frequently.Objective: To explore the effects ofWu Bie oral liquid on mice with immune premature ovarian failure (POF).Materials and methods: ZP3 were purchased from Chinese Peptide Company.Balb/c female mice model of POF was established by multiple sites subcutaneous injection of the mouse zona pellucida 3, and given daily Wu Bie oral liquid (16.5 g/kg) for 4 weeks after established.The AzpAb were determined by enzyme-linked immunosorbent assay, and T lymphocyte cell subsets of the spleen were detected by immunohistochemistry.The fas and fasL protein expression of the ovarian were detected by immunohistochemistry.The pathological changes of the ovary and spleen were analyzed.Results: Wu Bie oral liquid administered to mice failed to detect LD50, the mice maximum dose of the experimental results show that no significant toxicity in 20 mice after administration of 2 weeks, no death.Therefore, Wu Bie oral liquid is a safe drug.We found the increased AzpAb (8.74±0.99) in peripheral blood and CD4+/CD8+ (1.74±0.3 1) in the spleen, decreased Fas protein expression (0.54±0.29) and increased Fas-L protein expression (2.25±0.0) in the ovary were found in mice of POF model.Compared with the model group, the concentration of AzpAb (7.34±1.21) and CD4+/CD8+ (1.074±0.14) of Wu Bie oral liquid treated mice were significantly decreased (P<0.05).Moreover, Wu Bie oral liquid treatement induced significantly lower of Fas-L protein (1.29±0.86) expression than that in model group (p <0.05).Discussion and conclusion: The present study reveals the Wu Bie oral liquid can significantly decrease the concentration of AzpAb in peripheral blood, and regulate CD4+/CD8+ and the Fas/FasL system of mice with immune POF.
