With the advantages of high peak capacity, high resolution and high sensitivity, capillary isoelectric focusing (CIEF) has become a popular method for the separation ofbiomolecules.Although carrier ampholytes (CAs) are indispensable to establish a stable pH gradient in CIEF, the addition of CAs in the buffer might interfere the UV detection, especially at low wavelengths.In addition, they also deteriorate the detection of MS, making the direct hyphenation of CIEF and MS impossible.Therefore, recently many efforts have been made to remove or reduce the moving CAs in the buffer.In our previous study, such a problem could be solved by chemically bonding CAs on a monolithic column to form monolith with immobilized pH gradient (M-IPG) [1-3].
