Dragon' blood is widely used as a famous traditional Chinese medicine which has been found antimicrobial, antiviral, antitumor, antihemorrhagic, immunomodulatory, antiulcer, analgesic, antioxidative, anti-inflammatory, antimutagenic, and wound healing activity.Dracaena cochinchinensis (Lour.) S.C.Chen is one of the important sources of dragon's blood which distributes in south China, Vietnam and Laos.We have established a comprehensive two-dimensional PR/PRLC-UV separation system for the separation of Dracaena cochinchinensis (Lour.) S.C.Chen xylem containing resin (RX) [DOI: 10.1007/s 10337-011-2065-8].The system showed high peak capacity, sensitivity and separation efficiency.But there were some problems to be further improved: (1) by using trap mass spectrometry as the detector, more peaks can be detected and identified.(2) by using smaller C18-column to reduce the analysis time of the 2-D LC system.In the developed method, a CN-column was used in the first dimensional chromatography and a C18-column was used for second dimensional separation.About 80 components were detected in the RX extract with the two-dimensional RP/RPLC system.At same time 55 components were detected in the normal xylem extract.And 11 of the components were preliminarily identified.
