TrkA receptor signaling plays important role in NGF-induced neuronal survival and neurite outgrowth, so the trafficking of TrkA is essential for NGF-mediated signaling, but the molecular events that regulate this process is so far little known.In the current study, we identified syntaxin 8, a t-SNARE protein (target soluble N-ethylmaleimide-sensitive factor attachment protein receptor) as a novel TrkA-binding protein and overexpression and knockdown studies showed that syntaxin8 could significantly regulated TrkA surface expression and NGF indueed-neurite outgrowth in both TrkA-expressing PC12 cells and DRG neurons.Syntaxin8 was found involve in TrkA post-synthetic trafficking but not endocytic pathway.Our results suggest that syntaxin8 is a novel regulator of TrkA receptor trafficking.
