【目的】观察三七总皂苷(PNS)对类风湿关节炎(RA)患者环瓜氨酸肽(CCP)抗原特异性T细胞(CCP/AST)体外增殖及分泌干扰素-γ(INF-γ)的影响。【方法】体外分离培养24例RA患者外周血淋巴细胞,分为空白对照组、CCP组、甲氨喋呤(MTX)对照组及PNS组。体外培养72 h后,采用CCK-8法测定T细胞增殖水平及酶联免疫吸附法(ELISA)检测细胞培养上清中IFN-γ水平。【结果】CCP组患者外周血淋巴细胞增殖,与空白对照组比较明显升高,差异有显著性意义(P<0.01);PNS组与CCP组比较,淋巴细胞增殖可被明显抑制,差异有显著性意义(P<0.01);同时与空白对照组比较,CCP组的IFN-γ水平升高(P<0.05),其IFN-γ值经三七总皂苷作用后,IFN-γ水平有所下降(P<0.05)。【结论】三七总皂苷对CCP/AST增殖及IFN-γ分泌的抑制作用可能在类风湿关节炎的治疗中发挥重要作用。
【Objective】 To observe the effects of Panax Notoginseng Saponins (PNS) on the proliferation and secretion of IFN-γ (INF-γ) in vitro of CCP antigen-specific T cells (CCP / AST) in patients with rheumatoid arthritis )Impact. 【Methods】 Peripheral blood lymphocytes were isolated and cultured from 24 RA patients in vitro and divided into blank control group, CCP group, methotrexate (MTX) control group and PNS group. After cultured in vitro for 72 h, the proliferation of T cells was measured by CCK-8 assay and the level of IFN-γ in the cell culture supernatants was detected by enzyme-linked immunosorbent assay (ELISA). 【Results】 The proliferation of peripheral blood lymphocytes in CCP group was significantly higher than that in blank control group (P <0.01). Compared with CCP group, the proliferation of lymphocytes in PNS group was significantly inhibited (P <0.01). Compared with the blank control group, the level of IFN-γ in CCP group increased (P <0.05), and the IFN-γ level of IFN-γ decreased after treated with Panax notoginseng (P <0.05). 【Conclusion】 The inhibitory effect of Panax notoginseng saponins on CCP / AST proliferation and IFN-γ secretion may play an important role in the treatment of rheumatoid arthritis.
