25 microsatellite loci characterization and parent assignment of captive red pandas in Chengdu Resea

  The red panda (Ailurus fulgens) is one of the flagship species in worldwide conservation.Though the international lineage of red pandas in captivity was established in 1980,management of the captive population in China was haphazard,poorly documented,and characterized by frequent full-sib mating.These issues make the conservation and management of captive red pandas problematic.The microsatellite is a useful tool for determining parentage and evaluating population genetic diversity.So far,we characterized 25 microsatellite loci from two genomic DNA-enriched libraries of the red panda.The number of observed alleles among 35 samples of red pandas ranged from five to 17.Observed and expected heterozygosities were 0.286 to 0.971 and 0.121 – 0.909,and the mean polymorphic information content was 0.716.All loci followed Hardy-Weinberg expectations except two,which may due to the presence null alleles.Three pairs of loci exhibited significant linkage disequilibrium after Bonferroni correction for multiple comparisons.Then,we conducted the parent assignment of 32 red pandas at the Chengdu Research Base of Giant Panda Breeding (Sichuan,China).The results showed that all parents-offspring relationships are all confirmed.And there are high inbreeding coefficients in this population.

筛选出适合丹顶鹤遗传分析的微卫星位点对于丹顶鹤野生种群的保护遗传学研究以及人工散养丹顶鹤种群的遗传管理是非常重要的。本研究通过磁珠富集的方法分离丹顶鹤的微卫星分子标记。设计获得41对微卫星引物,选取合成39对。初步验证,有12对能够进行稳定的特异性扩增,并显示出多态性。此外本研究还从其他鹤类物种已经筛选的引物中筛选出在丹顶鹤物种中稳定扩增的6对引物。本次研究共获得18对丹顶鹤微卫星引物,为今后的丹
