Objective: To investigate the efficiency of human embryonic stem cells (hESC) induced into keratinocytes in vitro.Methods: The hESCs were divided into 6 groups: group A was induced by bone morphogenetic protein 4 (BMP4) and ascorbic acid (AA) via EB; group B was induced by BMP4, AA and retinoic acid (RA) via EB; group C was induced by KSFM via EB; group D was induced directly by BPM4 and AA; group E was induced directly by BMP4, AA and RA;group F was induced directly by KSFM.After 7 days of induction, moved each group of cell into new dishs and cultured by KSFM for another 4 weeks.Detected the differentiation markers (CK14, CK15, CK18, OCT4, P63) by fluorescence microscopy, flow cytometer and Realtime PCR after 35 days of differentiation.
