A New Expression Platform for Pharmaceutical Proteins and Peptides-Rice Endosperm Expression System

  To produce the safety and the cheaper pharmaceutical proteins and peptides for human Medicare,we developed a novel expressing platform using rice endosperm cell to highly express pharmaceutical protein and peptides.This platform uses an endosperm-specific prompter and signal peptide targeting the recombinant protein to the protein bodies,using optimized genetic codons and a fusion strategy to improve pharmaceutical proteins and small peptides expression in rice endosperm.Two proteins of recombinant human serum albumin (rHSA),human granular macrophage cell stimulate factor (GM-CSF) and a small peptide,human insulin-like growth factor-l(IGF-1) were tested.The results indicated that the expression level in rice endosperm were up to 108 μg/grain (rHSA) and 10μg per seed (GM-CSF) and 153 μg per seed (IGF-1).The data indicated that rHSA expressed in this platform processes proper folding and similar biochemical characters and biological activities to the native protein.Those demonstrated that this platform has the advantages of high protein accumulation,rapidly scale up,free of human pathogens,simple processing procedures and environment-friendly over other systems.So this expression platform could widely used for produce pharmaceutical proteins and peptides.
