Epilepsy is a common disorder in department of neurology,whose therapy has relied on anti-epilepsy drugs(AEDs)until now.However,over 30%patients failed to control the epileptic seizure by general AEDs.Crocin is one of major chemical components in Crocus sativus L.,a traditional Chinese medicine.Recently,it is reported that it exists a wide range of pharmacological activities,including anti-tumor,anti-depression,anti-anxiety,treatment of cardiac and cerebral ischemia,improve the cognitive function of neurodegenerative diseases and sedative hypnotic,etc.But the effect of crocin on epilepsy is still unclear.In hippocampal fast-kindling epilepsy model of mice,the present study evaluated the anticonsulvant effect of crocin.We have demonstrated that crocin(10 and 20mg/kg)increased the number of stimulations to reach fully-kindled stage and shorten the afterdischarge durations(ADDs)compared with control.Moreover,the animals rapidly reached the fully-kindled stage by several stimulations after withdrawal of the drug.We also tested the anticonvulsant effect of crocin on general seizures(GS)in fully-kindled mice.The results suggested that crocin(100mg/kg)decreased the incidence of GS and seizure severity in fullykindled mice compared with control.It even had a tendency of shortening generalized seizure durations and ADDs as well.In conclusion,we found that low dose crocin retarded the progression in hippocampal fast-kindling acquisition in mice,while high dose crocin relieved the GS in the fully-kindled mice.These indicate that crocin may be a potential anti-epilepsy compound and its mechanism is valuable with further researches.
