Toll样受体3在氧诱导的视网膜病变的大鼠模型中的作用研究

目的 探讨Toll样受体3(TLR3)在氧诱导的视网膜病变(OIR)的大鼠模型的作用.方法 新出生的SD大鼠暴露于80 %-21 %氧浓度中14天,然后在正常氧浓度条件下继续喂养4天.室内空气下喂养的正常大鼠作为对照组.两个实验组大鼠分别在出生17天后接受玻璃体腔内注射poly I:C和盐溶液.定量PCR分别检测OIR大鼠视网膜中的TLR3基因以及细胞因子TNF-α,IL-6的表达水平.免疫印迹学检测TLR3以及NF-κB的蛋白表达水平.结果 OIR大鼠视网膜中TLR3以及NF-κB的表达水平较正常对照组显著增加.OIR大鼠视网膜中细胞因子IL-6和TNF-α的基因表达也显著高于正常对照组.poly I:C能显著刺激NF-κB和细胞因子IL-6和TNF-α的表达水平.TLR3和细胞因子IL-6/TNF-α在OIR盐溶液注射组和正常poly I:C刺激组之间没有明显统计学差异.结论 TLR3信号通路在氧诱导的视网膜病变(OIR)大鼠模型的促炎症反应中发挥重要作用.
