Prevention ofIntra-abdminal Adhesion with Electrospinning PEG/PLGA Nanofibrous Membranes

  Postoperative adhesion formation is the prime complication that could happen after acute abdomen surgery.The application of physical barrier membranes at the operative site is considered as a potential method to solve this problem by preventing adhesive fibrotic tissue permeation.In this study,we fabricated electrospinning composite poly(ethylene glycol)(PEG)/Poly(lactic-co-glycolic)(PLGA)nanofibrous membrane to prevent abdomen adhesion,which could act as a barrier between the cecum and surrounding tissues,without interrupting mass transfer and cecum healing.PEG/PLGA nanofibrous membrane of 0%PEG(P0),5%PEG(P1),10%PEG(P2),15%PEG(P3),20%PEG(P4)and 25%PEG(P5)were prepared and characterized for physicochemical properties.The P0 membrane shows the best thermostability while P1 membrane exhibited the most homogenous morphology,the narrowest diameter distribution and the largest ultimate stress and strain.The P5 membranes had the lowest water contact angle.In vitro cell adhesion and proliferation tests with fibroblasts indicate that all nanofibrous membranes could inhibited the cell proliferation,P1 membranes with having the least cell attachment.In vivo application of nanofibrous membranes on the repaired site of rat cecum model demonstrated all of the membranes have certain effect in prevention adhesion formation.From gross observation,histological analysis and functional assays,we concluded that P1 membranes could function as an effective anti-adhesion membrane after cecum surgery in aclinical setting.
