Lead pollution severely endangers the environment and human health.Traditional measurements require expensive apparatuses and skilled personnel.Rapid,sensitive and portable detection of low level of Pb2+ in biological and environmental samples is of great importance to many applications.Herein,we proposed to utilize Pb2+ responsive DNAzyme crosslinked hydrogel for rapid,sensitive and portable detection of Pb2+.Polyacrylamide-DNA conjugates were crosslinked via Pb2+ DNAzyme to form the hydrogel.Gold nanoparticles(AuNPs)were encapsulated in the hydrogel as an indicator.After Pb2+ recognition by the enzyme strand,the cleaving of substrate crosslinkers occurred,inducing the release of AuNPs to be monitored by colorimetry.As low as 10 nM Pb2+ can be detected by naked eyes.Furthermore,to realize quantitative visual detection,the encapsulated AuNPs were replaced by gold-platinum nanoparticles(Au@PtNPs)with a portable volumetric bar-chart chip(V-Chip)device.The releasing Au@PtNPs from the hydrogel can efficiently catalyze the decomposition of H702 to generate a large volume of 02 to push the movement of an ink bar in the V-Chip for Pb2+ quantification.The limit of detection reached 2.6 nM.The present study will discuss the optimization of enzyme strand,synthesis of hydrogel,colorimetric and quantitative detection of Pb2+ by the V-Chip.
