Hematopoietic stem and progenitor cells (HSPCs) can be identified in Bone marrow (BM),umbilical cord blood and peripheral blood,but the current methods to harvest HSPCs are not satisfactory.We developed a new method to harvest HSPCs by implanting biomaterials into the spatium intermusculare of mice hind limbs,and detected the presence of HSPCs in the migrating cells (MCs) by phenotypically probing with CD34+Sca-1 + and functionally confirming the presence using colony-forming cell assay and assessing the long-term reconstitution ability after MCs transplantation,the population of CD34+Sca-1+ cells and amount of colony formation was much more in the MCs than the BM.In addition,transplanted MCs were able to home to the BM,muscle and spleen and induced a high efficiency long-term reconstitution in hematopoiesis in vivo than BM (>12 weeks).
