以聚合物载药胶束体系为例,采用新的表征聚合物结构特征的嵌段单元自相关(BUA)描述符的计算方法,建立了最佳结构-性能关系模型群。模型经内外检验参数评估、应用域定义,确认了模型的可靠性,确保了BUA描述符对载药机制的合理解释。聚合物中拨基氧的亲电作用对阿霉素载药量的影响最大,亲电作用越强,阿霉素的载药量越大,反映在嵌段单元上是“CL”越多,阿霉素载药量越大;对于星型聚合物,增加“PCL”嵌段长度或减小“PPEGMA”嵌段长度,都有利于聚合物胶束阿霉素载药量的增加;增加星型杂臂聚合物的"PDEAEMA”嵌段长度有利于载药量增加,而对于均臂聚合物则不利于载药量增加。通过对模型中BUR描述符的分析和机理阐释,进一步阐释了聚合物嵌段单元结构和拓扑结构对阿霉素载药量的影响。研究显示,BUA描述符能很好的在模型中反映聚合物的嵌段结构和拓扑结构对载药量的影响,因此在设计目标聚合物时,可以根据目前需求调整分子结构,也可以通过模型预测载药量。该研究为定量设计和开发合适载药量的聚合物及其胶束提供一种新的思路和理论方法,有望指导其它复杂结构化产品的设计和开发。
