Background: As one of the most important post-transcriptional modifications, alternative splicing is the key mechanism to expand transcript and protein diversity.Several genome-wide analyses also indicate that alternative splicing occurs frequently in the human genomes.One expression form of the alternative splicing is alternative promoter, and genes with alternative promoters are widely found in mammalian genomes.The epigenetics influence almost all the genes.Recent provocative studies point to some influence of DNA methylation in alternative splicing regulation.Methods: The sequences date of promoters were obtained from the TSSDB, and there were 61,168 constitutive promoters (CPs) and 40,268 alternative promoters (APs) from 76,485 human genes.Methylation date was downloaded from MethDB.The proportion of the 16 dinucleotides, GC-content, and the DNA information content of APs and CPs were calculated .The average values of the methylation occuring whether or not, which contains all the sites at every promoter sequences, was analysed.Results and discussion: There were no significant different in the 16 dinucleotides betwen APs and CPs.The GC-content of APs was higher than CPs.The methylation of APs is higher than CPs, except APs with two promoters and three promoters are lower than CPs.In the %99 confidence level, the distribution of the single nucleotides in APs and CPs sequences isnt irregular, two adjacent nucleotides exist some connections, precisely.Whats more, the two indexes of APs are higher than CPs .
