表观遗传学是分子遗传学研究的一个热点,DNA甲基化作为细胞记忆的一种机制,在维持正常细胞功能、遗传印记以及肿瘤发生中发挥着重要作用,是表观遗传学的核心领域之一。本文概述了DNA甲基化的相关机制,其中包括DNA甲基化模式特点及参与其改变与维持的酶类,同时对DNA甲基化分析的方法,即高效液相色谱法(High performance liquid chromatography, HPLC)、甲基化敏感的扩增片段长度多态性法(Metyhlation sensitive amplified fragment length polymorphism,MSAP)、亚硫酸氢盐测序法(bisulfate mapping)以及甲基化芯片法(DNA methylation chip technology)进行详尽的介绍和讨论。
