目的:建立同时测定三多丸中葛根素和芒柄花素含量的方法。方法:运用HPLC法进行测定研究,采用Agilent Hypersil C18(250 mm×4.6 mm,5μm)柱,以乙腈-水为流动相,梯度洗脱,检测波长为250 nm,流速1.0 mL/min。结果:HPLC法分析结果显示,葛根素和芒柄花素的色谱峰形好,分离度好,杂质无干扰,葛根素在0.288~2.88μg内呈现良好的线性关系,平均加样回收率为98.77%(RSD=1.2%),r=0.9999;芒柄花素在0.013~0.260μg内呈现良好的线性关系,平均加样回收率为97.68%(RSD=1.9%),r=0.9998。结论:该方法分离度高、专属性强、重现性好,可有效用于三多丸中葛根素和芒柄花素的含量测定。
Objective: To establish a method for simultaneous determination of puerarin and formononetin in Sanduo Pills. Methods: The HPLC method was used for the determination. The Agilent Hypersil C18 (250 mm×4.6 mm, 5 μm) column was used. The mobile phase consisted of acetonitrile-water with gradient elution. The detection wavelength was 250 nm and the flow rate was 1.0 mL/min. Results: The results of HPLC analysis showed that the peak shape of puerarin and formononetin were good, the degree of separation was good, the impurities had no interference, and the puerarin showed a good linear relationship in the range of 0.288 to 2.88 μg. The average recovery was 98.77. % (RSD = 1.2%), r = 0.9999; Formononetin showed a good linear relationship in 0.013 ~ 0.260μg, the average recovery was 97.68% (RSD = 1.9%), r = 0.9998. Conclusion: The method has high resolution, strong specificity and good reproducibility. It can be effectively used in the determination of puerarin and formononetin in Sanduo Pills.
