Hepatitis B virus (HBV) infection in the body primarily interferes with the functions of the liver by replicating in liver cells.The HBx protein of HBV is a small transactivator that is essential for infection by HBV and plays an important role in HBV pathogenesis related with liver diseases including hepatocellular carcinoma (HCC).HBx activates the RAS-RAF-MAPK pathway and enhances gene transcription from AP-1 that is heterodimer of c-Jun and c-Fos,which,in turn,leads to the proliferation of HBV and the transformation of hepatocyte.Our study shows that Mangiferin can obviously inhibit the activity of c-Jun N-terminal kinases (JNKs) of MAPK signal pathway in HBV-DNA transfected hepatocarcinoma cell(2215cell),prevent the formation of AP-1,which reveals that inhibiting the JNK/MAPK signal transduction pathway maybe one mechanism of mangiferin to inhibit the proliferation of HBV and prevent the transformation of hepatocyte.
