柠檬醛对指状青霉菌丝体转录组的影响

  指状青霉(Penicillium digitatum)引起的柑橘绿霉病可导致柑橘30-80%采后损失.前期结果表明,柠檬醛能显著抑制指状青霉菌丝体的生长,最小抑菌浓度(minimuminhibitory concentration,MIC)为2.0 μL/mL.为探讨柠檬醛抑制指状青霉菌丝体生长的分子机制,对其转录组进行了RNA-seq 分析.结果 表明,经1/2MIC 柠檬醛处理30min 后,共鉴定得到6335 个差异表达基因,包括2322 个上调基因和4033 个下调基因;这些基因可注释到155 个KEGG 通路,主要包括ABC 转运、糖酵解/糖异生、三羧酸循环、氧化磷酸化、硫代谢、氮代谢、磷酸肌醇代谢、脂肪酸生物合成、不饱和脂肪酸生物合成、脂肪酸代谢和类固醇生物合成、mRNA 监视、RNA 聚合酶、RNA 转运和氨酰tRNA 生物合成等代谢途径.其中,柠檬醛处理可诱导5 个麦角固醇生物合成途径相关基因(ERG7、ERG11、ERG6、ERG3和ERG5)表达下调,与麦角固醇以及羊毛甾醇的含量变化相符.此外,荧光定量PCR 结果显示,ERG11(羊毛甾醇-14α-脱甲基酶)为其中的关键调节基因.本研究结果表明,柠檬醛可通过影响指状青霉麦角固醇生物合成来抑制指状青霉生长.
