目的:研究降糖消渴颗粒含药血清(JGDS)对骨骼肌细胞胰岛素抵抗模型葡萄糖消耗量及葡萄糖摄取率的影响。方法:JGDS处理棕榈酸、小牛血清白蛋白(BSA)诱导而致胰岛素抵抗(IR)的C2C12骨骼肌细胞;通过检测培养液中的葡萄糖含量反映葡萄糖消耗量;2-NBDG葡萄糖摄入法观察细胞对葡萄糖的摄取率。结果:0.25mmol/L棕榈酸和1%BSA培养16h,使得C2C12骨骼肌细胞葡萄糖消耗量与葡萄糖摄取率显著下降(P<0.05),造成骨骼肌细胞IR模型,JGDS干预可使IR模型骨骼肌细胞葡萄糖消耗量显著增加,与正常血清相比,差异具有统计学意义(P<0.05),且该作用呈现一定的剂量依赖性;JGDS可增加IR骨骼肌细胞模型葡萄糖转运率,与模型组比较,差异具有统计学意义(P<0.05)。结论:降糖消渴颗粒含药血清能显著增加IR的C2C12骨骼肌细胞的葡萄糖消耗和摄取,这可能是降糖消渴颗粒改善骨骼肌IR的机制之一。
Objective: To study the effects of JGDS on glucose consumption and glucose uptake in skeletal muscle insulin resistance model. Conclusion: Jiangtangxiemai granule-containing serum can significantly increase the glucose consumption and uptake of IR C2C12 skeletal muscle cells, which may be one of the mechanisms of Jiangtangxueke granule in improving skeletal muscle IR.
