The success of the DNA chips for DNA-Directed Immobilization of proteins not only depends on the chemistry used for the immobilization of the oligonucleotides but also depends on the good accessibility and functionality of the surface-bound oligonucleotides, the density of attachment, and the reproducibility of the attachment chemistry.A general problem of protein biochips however, concerns their preparation by stepwise, robotic immobilization of multiple proteins at chemically activated surfaces, which is often obstructed by the intrinsic instability of many proteins and, thus, a loss of functionality.To overcome these problems, we have developed the DNA-directed immobilization (DDI) of proteins.
