目的 Ghrelin是生长激素促分泌素受体(GHSR)的内源性配体,在体内发挥多种生理学作用.近期的研究发现Ghrelin可促进小鼠成骨细胞(MC3T3-E1)增殖和分化,抑制MC3T3-E1凋亡,但具体机制尚不明确.本文旨在研究Ghrelin对MC3T3-E1凋亡的影响及其作用机制.方法 以10-9 ~ 10-11MGhrelin干预MC3T3-E1,分别以TUNEL法和ELISA法检测其细胞凋亡情况;Western blot法检测Bcl-2、Bax蛋白表达以及caspase-3的分泌;RT-PCR和Western blot法检测GHSR在MC3T3-E1的表达;使用RNA干扰技术阻断GHSR表达.
