目的:研究黄芩水提液干预对呼吸道合胞病毒(RSV)感染BALB/c小鼠血浆和肺组织脂质代谢异常的调节作用。方法:RSV滴鼻感染BALB/c小鼠,黄芩水提液灌胃给药进行干预,采集血浆和肺组织样品进行基于超高效液相-二维线性离子阱高分辨静电场组合质谱仪(UPLC-LTQ/Orbitrap XL)的脂质组学研究,考察干预前后脂质代谢物组的变化,寻找潜在生物标志物,探讨黄芩水提液抗病毒作用的途径。结果:正常组、模型组、利巴韦林组、黄芩组血浆和肺组织脂质谱均得到明显分离,在血浆和肺组织中分别鉴定了与RSV感染相关的9种和7种生物标志物,主要包括甘油磷脂酰胆碱、鞘磷脂和甘油三酯,黄芩水提液干预对部分异常脂质代谢具有明显的改善作用。结论:黄芩水提液能有效缓解RSV感染所致的甘油磷脂酰胆碱、鞘磷脂和甘油三酯代谢紊乱,表明黄芩抗病毒的作用机制可能与调节这些代谢途径有关。
OBJECTIVE: To study the regulatory effect of aqueous extract from Scutellaria baicalensis Georgi on lipid metabolism in plasma and lung tissue of BALB / c mice infected with respiratory syncytial virus (RSV). Methods: BALB / c mice were inoculated intranasally with RSV, and the aqueous extract of Scutellaria baicalensis Georgi was administrated by intragastric administration. Plasma and lung tissue samples were collected for determination by UPLC (High Performance Liquid Chromatography - Two Dimensional Linear High Performance Ion Trap Ion Trap Mass Spectrometry) -LTQ / Orbitrap XL) to investigate the changes of lipid metabolites before and after the intervention, searching for potential biomarkers and exploring the anti-viral effect of Scutellariae Radix extract. Results: The lipid profiles of plasma and lung tissues of normal group, model group, ribavirin group and Scutellaria baicalensis Georgi were significantly separated, and 9 and 7 biomarkers related to RSV infection were identified respectively in plasma and lung tissue. Mainly including glycerophosphatidylcholine, sphingomyelin and triglycerides, skullcap extract on some abnormal lipid metabolism has a significant improvement. Conclusion: Scutellariae water extract can effectively alleviate the metabolism of glycerol phosphatidylcholine, sphingomyelin and triglycerides caused by RSV infection, indicating that the anti-virus mechanism of Scutellaria baicalensis may be related to the regulation of these metabolic pathways.
