Transcription activator-like effector nucleases(TALENs)is a newly developed powerful tool for gene editing,and it have been applied to various organisms.We have successfully developed rice minichromosomes system by telomere-mediated chromosomal truncation method in our previous study.Definitely,the minichromosomes system could be improved when combined with TALENs.In this study,three pairs of TALENs(TALENs-1,TALENs-2,TALENs-3)were designed targeting different sites in a gus gene in rice minichromosomes line 1004-011.These TALENs pairs were transformed into 1004-011 callus respectively through biolistic delivery system.The transformants were detected by GUS-staining assay.Preliminary data shows that 1 out of 17 TALENs-3 transformants showed no β-glucuronidase activity,indicating that the gus gene may be interrupted by TALENs.While TALENs-1 and TALENs-2 transformants were all GUS positive,indicating that the gus gene was not affected by these TALENs.
