The pharmacokinetics and tissue distribution of harmine were investigated in mice after oral administration at dose of 40 mg/kg using RP-HPLC.Blood and tissues samples were taken at 0.083, 0.17、 0.33, 0.5, 0.75, 1,1.5, 2, 3, 4, 6, 8 and 12 h after single doses of 40 mg/kg harmine.Blood and tissues samples were taken to investigate harmine accumulation in mice at 1 h after once-a-day dosing of 40mg/kg harmine over 21 consecutive days.The maximum harmine blood/tissues concentrations (Cmax) were observed within 0.75 h after a single dose of 40mg/kg harmine.The mean Cmax and mean AUC0-∞ of harmine in different tissues were 3-to 111-fold and 3.3-to 32-fold higher than that in blood, respectively.The highest AUC value was found in stomach and intestines with values of 210.5 and 185.2 (μg·h/g), respectively.The AUC values were 84.60,45.55 and 38.0 (μg·h/g) in thymus, spleen, and liver, respectively.
