Objective:Weexpecttofindsomelymphomaassociatedmoleculesthroughfundamentalresearchesandwanttodevelopthem tobediagnosticmolecularprobesinthefutureandappliedtoclinicaldiagnosisandtreatment. Materials and Methods: BCMAandRasGRP4wasfoundbygenechipastargetmolecules,varioustypesandmalignantlevelsof largeBlymphoma,follicularlymphoma,Burkittlymphoma,mucosaassociatedlymphoidtissue,smalllymphomaandplasmacell lymphomaandsoonwereallcollectedandthesecaseswereallconfirmedbypathology.VerifytheexpressionofBCMAby immunohistochemistryandatthesametime,confirmitseffectsonlymphomacellsapoptosisbyelectroporationof correspondingcDNAintheRajicellline.VerifytheexpressionofRasGRP4bythesameimmunohistochemistrymethod,andthe shRNAofRasGRP4weretransfectedintoSUDHL-4celllinebyelectroporationtostudyitsinfluenceonapoptosisofthediffuse largeBcelllymphomacells.Atthesametime,Collected14casesofdiffuselargeBlymphomawhichwasconfirmedbypathology, andBCMAandRasGRP4moleculesweredetectedbyimmunohistochemistryinthesamecondition.Throughthestatistical analysis,respectivelyexplorethecorrelationbetweentheSUVmaxofPET-CTandtheexpressionofBCMAandRasGRP4. Results:29casesoflymphomaand5casesofnormalcontrolswerestainedbyimmunohistochemistryandtheexpressionof BCMAwerelowerthanthatofnormalcontrolgroupinlargeBlymphoma,follicularlymphoma,Burkittlymphoma,mucosa associatedlymphoidtissue,smalllymphomaandplasmacelllymphoma,atthesametime,indifferentmalignantlevelsof follicularlymphoma,theexpressionofBCMAwereassociatedwiththemalignantlevel,whenmalignantlevelwashigher,lower BCMAexpressionwasobserved.AfterthetransfectionofthecDNAofBCMAorshRNAOFRasGRP4intheRajicellline,theearly stageandlatestageapoptosisrateoftransfectiongroupwerequitehigherthanthecontrolgroup.14casesofdiffuselargeB lymphomapatientswithanaverageageof63yearsoldwereincludedand9caseswerefemale,5weremale.Amongthem, SUVmaxof9caseswereobtainedatlymphnodelesions,theremaining5casesoccurinthebrain,frontalsinus,tonsil,thoracic vertebraandmediastinum.Amongallincludedcases,themaximumvalueofSUVmaxwas51,whichlocatedinthetonsilandthe minimumwas12.3andlocatedinthelymphnode,theaveragevalueofSUVmaxofallincludedpatientswere22.4.Itisfoundthat therewasnotaclearnegativerelationshipbetweenSUVmaxandtheIODofBCMA(r=0.0867,p=0.8245),whiletherewasa relationshipbetweenaverageIODofRasgrp4andpatientsSUVmaxvalue,correlationcoefficientwas0.73,thevalueofpwas 0.025,whichwasstatisticalsignificant. Conclusion:TheexpressedBCMAislowerthannormallymphnodesinavarietyoflymphomasubtypesandtheexpressionlevel ofBCMAmaybeabletoreflectlymphomagrade,whileexpressionofRasGRP4issignificantlyhigherinthediffuselargeBcell lymphomathannormallymphnodes.BCMAandRasGRP4moleculesmayplayanimportantroleinthecellproliferationand apoptosisresistancemechanisminthemalignantlymphoma.TheexpressionofRasGRP4wasassociatedwithSUVmaxvalueandit mayreflectmetabolicactivity,proliferationandmalignantinvasiveofdiffuselargeBcelllymphoma.
