  In recent years,field surveys showed that soybean propagation and plant virus transmission by Acyrthosiphon solani are on the rise.In order to probe into the interaction mechanism between propagation of mosaic virus(soybean mosaic virus,SMV)by means of Acyrthosiphon solani and its Buchnera-producing GroEL protein,we cloned the full-length Buchnera groEL sequence of Acyrthosiphon solani and analyzed the effects of expression levels of Buchnera groEL at different developmental stages.The full-length groEL sequence is 1647 bp encoding 548 amino acid residues with the predicted molecular weight 69 kDa from the Buchnera of Acyrthosiphon solani.Then the gene was recombined into pET21b and expressed in host bacteria(DE3).Analyzed the he changes of expression levels of groEL gene of Acyrthosiphon solani at different time and different developmental stages after feeding SMV by Real-time quantitative PCR technology and SPSS 17.0 statistical software(P<0.05).The Buchnera groEL gene expression levels was increased significantly with the extension of time of feeding SMV of Acyrthosiphon solani,the increased range between 1.35-5.61 times,and more higher in wingless and winged adults then the other four nymphs period.All these results above showed that the expression of groEL gene was induced in Acyrthosiphon solani after feeding on Soybean mosaic virus,ensure the SMV without degradation during transportation,which increases the possibility of Soybean mosaic virus spread in soybean field.
