Unimolecular micelle is a kind of polymeric nanoparticle that is free from disassembly under dilute condition,which is advantageous over traditional micelle in the context of therapeutic loading.Amphiphilic bottlebrushes are good candidates to form such kind of unimolecular micelle owing to their unique structures.We report here a new type of side-chain cleavable amphiphilic bottlebrushes that could be used as stimulus-responsive nanocarrier.These bottlebrushes were obtained by the ROMP of designed macromonomers.The macromonomers are amphiphilic and have a H2O2-responsive linker in the junction.They were synthesized by a combination of Passerini reaction and click chemistry.Four bottlebrushes different in side-chain density,side-chain length and backbone length were synthesized for comparison.We investigated their H2O2-responsive property via NMR,GPC,DLS,etc.Our work has demonstrated that ROMP-based bottlebrushes are novel unimolecular polymeric nanocarriers with tunable size,uniform chemical structure,narrow distribution and facile synthesis.
