目的:探讨溃疡性结肠炎(UC)大鼠肺损伤的血小板活化机制,观察中药干预作用。方法:免疫复合法复制UC大鼠模型,将大鼠随机分为7组,即正常组,模型组,西药组,从肺论治组,从肠论治组,苦参碱治疗组,鱼腥草素钠治疗组,分别于0周、4周后杀检取材,采用放射免疫法测定大鼠肺组织中血栓烷素B2(TXB2)、6-酮-前列腺素F1α(6-Keto-PGF1α)含量,ELISA法检测大鼠血清中P-选择素含量。结果:0周末模型组肺组织TXB2含量显著高于正常组(P<0.01),4周末从肺论治组与苦参碱治疗组TXB2值显著低于模型组(P<0.01);4周末苦参碱治疗组6-Keto-PGF1α含量较模型组和西药组明显升高(P<0.01,P<0.05);0周末和4周末模型组肺组织TXB2/6-Keto-PGF1α含量均较正常组显著升高(P<0.05);4周末各中药组TXB2/6-Keto-PGF1α含量较模型组显著降低(P<0.01),且从肺论治组和苦参碱治疗组TXB2/6-Keto-PGF1α含量较西药组显著下降(P<0.05)。0周和4周末,模型组血清P选择素含量均显著高于正常组(P<0.01,P<0.05)。结论:UC大鼠血小板的持续异常活化易导致肺组织局部微循环障碍,中药组在减轻肺内炎症和损伤方面具有优势。
Objective: To investigate the mechanism of platelet activation induced by lung injury in rats with ulcerative colitis (UC) and observe the effect of traditional Chinese medicine. Methods: The UC model was established by immunocomplex method. The rats were randomly divided into 7 groups: normal group, model group, western medicine group, lung treatment group, treatment group of Gutanzhi group, matrine group, Hormone sodium treatment group, respectively, at 0 and 4 weeks after the killing test material, using radioimmunoassay in rats lung tissue thromboxane B2 (TXB2), 6-keto-prostaglandin F1α (6-Keto-PGF1α) Content, P-selectin content in rat serum by ELISA. Results: The levels of TXB2 in the lung tissue of the model group at the end of the weekend were significantly higher than those of the normal group (P <0.01). The TXB2 value of the lung group and the matrine group was significantly lower than that of the model group at the end of the 4th week (P <0.01) The content of 6-Keto-PGF1α in the treatment group was significantly higher than that in the model group and the western medicine group (P <0.01, P <0.05). The content of TXB2 / 6-Keto-PGF1α in the model group at the 0th and the 4th week TXB2 / 6-Keto-PGF1α in TCM group was significantly lower than that in model group at 4 weeks (P <0.01), and TXB2 / 6-Keto PGF1α content significantly decreased compared with western medicine group (P <0.05). Serum P-selectin levels in model group were significantly higher than those in normal group at 0 and 4 weeks (P <0.01, P <0.05). CONCLUSION: Sustained abnormal activation of platelets in UC rats can lead to local microcirculation disturbance in lung tissue. TCM group has the advantage of reducing inflammation and injury in lung.
