Parkinsons disease (PD) is one of the most prevalent neurodegenerative diseases characterized by accumulation of α-synuclein (α-syn).Geniposide (Gde) is a major iridoid glycoside isolated from the fruit of Gardenia jasminoides Ellis., which has been widely used in traditional Chinese medicine. In this study, the novel function of Gde as a neuronal autophagy inducer was reported.Gde induced autophagy in different neuronal cell lines, including N2a,PC12 and SH-SY5Y cells. Moreover, Gde promoted autophagy in Drosophila fat body and in the brain of ICR mice. The neuroprotective effect of Gde was evaluated on Drosophila PD model overexpressing wild-type α-synuclein (WT-αsyn).Gde down-regulated the expression level of WT-αsyn in the brains of the flies. The motor function of the flies receiving Gde was improved. The loss of dopaminergic neurons was also ameliorated by Gde.Collectively, we have identified Gde as a neuronal autophagy inducer, which can promote the clearance of WT-αsyn and protect neurons.
